Abstract

Komina masline bogata je polifenolima koji imaju antioksidacijski učinak. Mogu djelovati protuupalno, smanjuju rizik od kardiovaskularnih bolesti, dijabetesa, osteoporoze, karcinoma i neurodegenerativnih bolesti. Nakon proizvodnje maslinovog ulja ostaje velika količina komine masline čije zbrinjavanje predstavlja ekološki problem. Najčešće predstavlja otpad, a ponekad se koristi kao gnojivo ili za proizvodnju energije. Ovaj diplomski rad izrađen je u svrhu ispitivanja antioksidacijskog učinka komine masline na model-sustavu liposoma gdje je ispitana inhibicija lipidne peroksidacije nativnog uzorka komine masline, uzoraka komine masline s ciklodekstrinima RAMEB (nasumično metilirani ciklodekstrin) i HPB (hidroksipropil-β-ciklodekstrin) kao ekscipijensima i BHA kao dokazanim antioksidansom. Optimizirani su uvjeti oksidacije u kojima se na model-sustavu liposoma može pratiti stvaranje MDA. Svi uzorci pokazali su inhibiciju lipidne peroksidacije, a najveću je pokazao uzorak s RAMEB ciklodekstrinom što ukazuje na protektivno djelovanje ciklodekstrina na polifenole komine masline. Komina masline, koja prethodno nije puno ispitivana, predstavlja potencijalan izvor sirovine za izolaciju prirodnih polifenola koji bi se mogli primijeniti u farmaceutskoj ili prehrambenoj industriji i zato ovaj rad predstavlja važan korak u istraživanju njezinih potencijala kako bi se u budućnosti ona mogla iskoristiti.

Abstract (english)

Olive pomace is rich in polyphenols, a substance which can have antioxidant and anti-inflammatory effect and also reduce the risk of cardiovascular diseases, diabetes, osteoporosis, carcinoma and neurodegenerative diseases. It ensues in large quantities as a result of the extraction of olive oil. However, the disposal of such large quantities of olive pomace creates enviromental problems. In most cases, olive pomace represents waste, but it can sometimes be used as fertilizer or as a material for energy production. This diploma paper aims to investigate the antioxidant effect of olive pomace on liposome model system which was used to examine the inhibition of lipid peroxidation of the original olive pomace sample, olive pomace and randomly methylated β-cyclodextrins (RAMEB) and hydroxypropyl β-cyclodextrins (HPB) sample, used as excipients, and β-hydroxyanisol (BHA) sample, used as a proven antioxidant. The conditions for oxidation have been optimized so the creation of malonaldehide (MDA) could be observed on liposome model system. All samples have shown the inhibition of lipid peroxidation. The sample consisting RAMEB has shown the greatest inhibition which may indicate the protective effect the cyclodextrin has on polyphenols in olive pomace. Therefore, olive pomace, which was not previously researched in greater detail, represents a potential source of raw material that can be used for the isolation of natural polyphenols, which may be utilized in pharmaceutical and food industry. Thus, this diploma paper represents an important step towards researching the potential of olive pomace and its future uses.
