Abstract

Dijabetes mellitus je metabolički poremećaj kompleksne etiologije okarakteriziran stanjem kronične hiperglikemije s poremećajem metabolizma ugljikohidrata, masti i proteina, što je rezultat nedostatka izlučivanja inzulina i/ili djelovanja inzulina na razini stanice. Jedan od načina regulacije postprandijalne glikemije je usporavanje razgradnje i apsorpcije ugljikohidrata unesenih prehranom, djelovanjem na enzim α-amilazu. Taj način liječenja trenutno je moguć akarbozom koja je inhibitor α-amilaze. U posljednje vrijeme proučavaju se potencijalni novi antidijabetici, bioaktivni spojevi izolirani iz biljaka. Primjena bioaktivnih komponenata sa antidijabetičkim učinkom bila bi osobito korisna kao sastavni dio dijetoterapije pacijenata sa preddijabetesom. Komina masline je čvrsti nusprodukt koji nastaje u proizvodnji maslinovog ulja, i sadrži polifenolne spojeve. U ovom radu ispitan je mogući inhibitorni učinak ekstrakata komine masline na aktivnost α-amilaze. U svrhu dodatnog poboljšanja učinkovitosti nativnog ekstrakta, pripremljeni su i istraženi uzorci inkapsulirani ciklodekstrinima. Sva tri korištena uzorka- nativni, HPB i RAMEB pokazali su inhibitorni učinak na α-amilazu. IC50 vrijednosti redom su iznosile 6.7, 4.5, i 3.2 mg/mL (akarboza 0.04 mg/mL). Ovim istraživanjem pokazano je da ekstrakt komine masline ostvaruje inhibitorni učinak na α-amilazu, tim bolji ako je uklopljen u ciklodekstrine. Ovi bi rezultati mogli poslužiti za daljnja istraživanja, prije svega potvrde i izolaciju spojeva koji su odgovorni za inhibitorni učinak, a potom i razvoj potencijalnih antidijabetika dobivenih iz ekstrakata komine masline.

Abstract (english)

Diabetes mellitus is a metabolic disease of complex etiology characterized by chronic hyperglycemia with disorder of carbohydrate, fat and protein metabolism resulting from defects in either insulin secretion or insulin action, or both. One approach for decreasing postprandial hyperglycemia is slowing down degradation and absorption of dietary carbohydrates, acting upon α-amylase. This approach for treating the disease is currently possible with acarbose which is well known α-amylase inhibitor. Recently, several researches have been investigating potential new anti-diabetic substances, which are byoactive compounds isolated from plants extracts. Use of these bioactive compounds with anti-diabetic activity would be particularly usefull as part of nutrition therapy for patients with pre-diabetes. Olive pomace is a solid by-product resulting from olive oil production, and it is rich source of polyphenols. In this study, possible inhibitory effect of olive pomace extracst on α-amylase activity has been investigated. All analyzed samples - native, HPB and RAMEB have shown inhibitory effect on α-amylase. Calculated IC50 values were 6.7, 4.5 and 3.2 mg/mL respectively (acarbose 0.04 mg/mL). This study proved that olive pomace extracts exhibited considerable α-amylase inhibiton activity, which was additionaly improved by cyclodextrin encapsulation. Obtained results could be used for further studies, firstly for investigation of active compounds responsible for observed activity, and subsequent develiopment of novel anti-diabetics isolated from olive pomace extracts.
