Abstract

Kolorektalni karcinom jest zloćudna novotvorina epitelnih stanica sluznice debelog crijeva.
Spada u skupinu tumora s najvećom incidencijom i visokom stopom smrtnosti.
Razvoj kolorektalnog karcinoma potiču stanične promjene genske ekspresije i sinteze proteina. Pogreške u genomu
matične stanice česte su kod kolorektalnog karcioma, a postoje tri različita aspekta genomske nestabilnosti: CIN, MSI
i putevi fenotipa CpG. Promjene se događaju na genima za proteine vezane za diobu matičnih stanica AURK i Plk.
Također vrlo česte su pogreške i na tumor supresorskim genima TP53, DCC i APC.
Osim gena za rast stanice narušeni su i menhanizmi za regulaciju stanične diferencijacije i stanične signalizacije, a
osobito Ras-Raf-MAPK puta. Proteinski biopokazatelji se najčešče koriste za otkrivanje i dijagnozu tumora debelog
crijeva, te praćenje tijeka bolesti, a najistaknutiji su CEA, CA 19-9, MMP9 i NGAL. Biopokazatelji olakšavaju
dijagnozu bolesti, propisivanje terapije i meta su novih terapijskih sustava.

Abstract (english)

Colorectal cancer is a malignant neoplasm of epithelial cells of the colon. It belongs to the tumor group with the
highest incidence and high mortality rate. The development of colorectal cancer promotes cellular gene expression and
protein synthesis. Genome defects are common in colorectal cancer, and there are three different aspects of genomic
instability: CIN, MSI and CpG phenotype pathways. Changes occur on protein genes related to the division of the
stem cells AURK and Plk. Also very common are faults in tumor suppressor genes TP53, DCC and APC. In addition
to cell growth genes, there are also maladies to regulate cell differentiation and cellular signaling, especially Ras-RafMAPK pathways. Protein biomarkers are most commonly used to detect and diagnose colon cancer and disease
tracking and CEA, CA 19-9, MMP9 and NGAL are most prominent. Biomarkers facilitate disease diagnosis,
prescribing therapy and targeting new therapeutic systems.
