Abstract

Svrha ovog rada bila je ispitati biokompatibilnost/citotoksičnost različitih tipova liposoma s
azitromicinom na MJ90hTERT fibroblastima. Konvencionalni, deformabilni,
propilenglikol i kationski liposomi s azitromicinom međusobno su se razlikovali po
(fosfo)lipidnom sastavu i prisustvu suotapala (propilenglikol). Liposomi su pripravljeni tzv.
film metodom, a primjenom ekstruzije postignuta je prikladna veličina liposoma za
topikalnu primjenu. Sastav liposoma utjecao je na fizikalna svojstva liposoma, posebice
naboj na površini i uspješnost uklapanja azitromicina. Provedena in vitro ispitivanja
citotoksičnosti pokazala su biokompatibilnost svih ispitivanih liposoma s fibroblastima u
koncentracijama azitromicina s kojima se postiže adekvatan antimikrobni učinak
liposomskih formulacija. Usporedba različitih tipova liposoma pokazala je najbolju
kompatibilnost propilenglikol liposoma, a najmanju kationskih liposoma koja je posljedica
prisutnosti kationskog lipida (DODAB-a) u fosfolipidnom dvosloju.

Abstract (english)

The aim of this study was to investigate the biocompatibility/cytotoxicity of the different
types of azithromycin-loaded liposomes on the MJ90hTERT fibroblasts. Conventional,
deformable, propylene glycol and cationic liposomes encapsulating azithromycin, and
differing in (phospho)lipid composition and the presence of co-solvent (propylene glycol),
were prepared using the film hydration method. The suitable size of the liposomes for topical
application was achieved by their extrusion through the membranes of 400 and 200 nm. The
composition of liposomes has proven to influence the physical properties of liposomes,
particularly the surface charge and the entrapment efficiency of azithromycin. All the
azithromycin-loaded liposomes were biocompatible with the fibroblasts at azithromycin
concentrations showing appropriate antibacterial effect. The comparison of the different types
of liposomes demonstrated the highest level of compatibility with the MJ90hTERT cells for
propylene glycol liposomes and the lowest for cationic liposomes, due to the presence of the
cationic lipid (DODAB) in the (phospho)lipid bilayers.
