Abstract
Proteini su jedne od najvažnijih makromolekula u ljudskome tijelu. Kontroliraju mehanizme koji dovode do različitih bolesti i zbog toga su sveprisutna tema istraživanja. Tema ovog rada je identifikacija proteina iz smjese metodom spektrometrije mase. U istraživanju je korišten MALDI-Q-TOF spektrometar mase. Uvođenje MALDI ionskog izvora spojenog sa tandem Q-TOF analizatorom je osiguralo sredstva za dobivanje pouzdanih MS i MS/MS podataka. Cilj je dobiti podatke o masama peptida i fragmenata koji su korisni za identifikaciju proteina i sastava smjese uz pomoć proteinskih baza sekvenci. U istraživanju je kao uzorak korištena ljudska plazma i identificiran albumin kao najobilniji protein.

Proteins are one of the most important macromolecules in the human body. They control the mechanisms causing various diseases and that makes them an ubiquitous research theme. Topic of this thesis is protein identification from mixture using mass spectrometry. MALDI-Q-TOF mass spectrometer was used in the research. Introduction of a MALDI ion source coupled to a tandem Q-TOF analyser has provided means to obtain reliable MS and MS/MS data. Aim is to acquire data about peptide masses and fragments useful for identification of proteins and mixture composition with the help of protein databases. Sample being used was human plasma and albumin was identified as the most abundant protein.
