Abstract
Industrijska konoplja (Cannabis sativa) je vrlo korisna biljka koja ima različite primjene u prehrani, graditeljstvu, tekstilnoj industriji itd. Kao i sve biljke podložna je kontaminaciji s plijesnima koje predstavljaju ekonomski i zdravstveni rizik. Zato je bitno eliminirati plijesni i utvrditi najbolji i najdjelotvorniji postupak mikrobiološke dekontaminacije. Jedan od jednostavnih, brzih, djelotvornih i sigurnih metoda mikrobiološke dekontaminacije je metoda pomoću γ-zračenja. Djelovanje zračenja na izloženu tvar dovodi do urušavanja genetičkog materijala stanica plijesni, oštećuje staničnu membranu i enzime što rezultira poremećajem normalne funkcije i naposljetku smrću stanica. Za odgovarajuću primjenu metode vodi se računa o primijenjenoj dozi kojom se mora postići željeni učinak zračenja, a da se pritom ne induciraju nikakve kemijske ili fizičke promjene u tvari. Ovaj rad se bavio istraživanjem dekontaminacije sjemenki industrijske konoplje od plijesni primjenom ionizirajućeg zračenja. Sjemenke industrijske konoplje su sterilizirane zračenjem te namjerno kontaminirane s određenim vrstama plijesni. Mikrobiološkom analizom sjemenki industrijske konoplje prethodno su određene plijesni Aspergillus flavus, Phoma spp., Fusarium spp. i Cladosporium spp. Pripravljene su suspenzije s koncentracijama od 102 spora/ml od svake vrste plijesni. Osim toga pripravljena je i suspenzija sa svim ovim vrstama isto ukupne koncentracije 102 spora/ml, a za kontrolni uzorak je korištena sterilna voda. Svakom uzorku od 20 g sjemenki konoplje je dodano 2 ml suspenzije i kontrolnom uzorku 2 ml sterilne vode. Nakon inkubacije uzorci su ozračeni s 10, 12 i 15 kGy zračenja te nasađeni na DRBC agar. Porast se kontrolirao sedmi, četrnaesti i dvadeset osmi dan. Rezultati su pokazali da primijenjene doze zračenja ne samo reduciraju broj plijesni, nego daju uzorke bez prisutnih plijesni tj. dobiveni su sterilni uzorci.

Industrial hemp (Cannabis sativa) is a very useful plant that has different applications in livelihood, architecture, textile industry etc. As any other plant it is susceptible to mold contamination and that represents an economic and a healthcare risk. That's why it is important to eleminate mold and detect the best and the most effective procedure of microbial decontamination. One of simple, fast, effective and methods of microbial decontamination is ă–irradiation. Action of ă–irradiation on exposed matter collapses the genetic material of the cell, damages cell wall and enzymes which results in disoreder of normal functions and cell death. For appropriate application of the method one must mind the applied dose which has to achieve the wanted radiation effect whilst not inducing chemical or phisical changes in the matter. This work explored decontamination from mold of industrial hemp seed by using ă–irradiation. Industrial hemp seed were sterilized using ă–irradiation and deliberately contaminated with certain species of molds. Aspergillus flavus, Phoma spp., Fusarium spp. and Cladosporium spp. were previously determined by microbial analyses of industrial hemp seed. Suspensions with concentrations of 102 spores/ml of every mentioned mold species were prepared. Beside that, a mixture of all four species with total concentration of 102 spores/ml was also prepared and for a control sample sterile water was used. 2 ml of suspension were added to samples of hemp seed each weighing 20 g and 2 ml of sterile water were added to the control sample. After incubation the samples were irradiated with doses of 10, 12 and 15 kGy and planted on DRBC agar plates. The growth was monitored on seventh, fourteenth and twentyeighth day. The resultst showed that applied radiation doses not only reduce the number of mold but also give sterile samples.
