Abstract
Liposomi zauzimaju istaknuto mjesto među terapijskim nanosustavima za parenteralnu primjenu i najduže su od svih nanosustava prisutni u kliničkoj praksi. Zbog svoje sličnosti s biološkim membranama, netoksičnosti, neimunogenosti i biorazgradljivosti liposomi su fiziološki prihvatljivi što im osigurava primjenu u različitim terapijskim područjima: onkologija, infektivna oboljenja (virusna, bakterijska, gljivična, parazitska), dijagnostika, hormonska terapija, vakcinacija i drugo. Uklapanjem u liposome mijenjaju se farmakokinetička svojstva lijeka i poboljšava njegova bioraspoloživost te omogućava ciljana dostava lijeka na oboljelo mjesto, smanjujući sistemske nuspojave i toksičnost. Ovaj rad donosi pregled istraživanja i kliničke primjene liposoma namijenjenih ciljanoj parenteralnoj terapiji prvenstveno tumorskih oboljenja, antimikrobnoj terapiji i vakcinaciji. Obrađeni su komercijalno dostupni liposomi za parenteralnu primjenu te oni koji se nalaze u kliničkim ispitivanjima.

Among different nanopharmaceuticals (nanosystems, nanocarriers) investigated for parenteral drug delivery liposomes have the longest history in clinical practice. Because of their structural similarity with biological membranes, biodegradability, biocompatibility, non-immunogenicity and non-toxicity, liposomes are physiologically acceptable and can be used in different therapeutic areas: oncology, infectious diseases (viral, bacterial, fungal, parasitic), diagnostics, hormone therapy, vaccination, etc. Liposomes are able to modify pharmacokinetic properties of the encapsulated drug and enhance its bioavailability. Moreover, they allow targeted drug delivery to the pathological area, thus reducing systemic side effects and toxicity. This thesis provides an overview of the research and clinical applications of liposomes intended for targeted drug delivery by parenteral route of drug administration, primarily tumor targeting, antimicrobial therapy and vaccination. Majority of the presented formulation of liposomes are commercially available or are in the final stages of clinical examinations.
