Abstract

Cilj ovog diplomskog rada bila je priprema mukoadhezivnih i mukopenetrativnih, polianhidridnih nanočestica s melatoninom. Melatonin je endogeni spoj koji je pokazao značajan terapijski potencijal u liječenju bolesti usne šupljine, stimulaciji imunološkog sustava te kao adjuvans u borbi protiv infekcija. Uklapanjem melatonina u nanočestice omogućava se njegova apsorpcija kroz mukozu te izbjegavanje prvog prolaska kroz metabolizam, brzi početak djelovanja i ciljano djelovanje. Oblaganjem površine nanočestica polietilenglikolom moţe se utjecati na njihov transport na sluznici te tako pripremiti mukoadhezivne (nePEGilirane) i mukopenetrativne (PEGilirane) nanočestice. Pri tome je nuţna optimizacija tehnološkog postupka pripreme s ciljem priprave nanočestica ciljanih svojstava. Rezultati ispitivanja pokazuju da promjenom koncentracije polianhidridnog polimera je moguće kontrolirati veličinu nanočestica, dok brzina dodatka vode i miješanje sustava tijekom postupka desolvatacije u pravilu pridonosi nastajanju monodisperznih nanočestica. Nanočestice ţeljenih svojstava pripremljene su pri brzini dodatka vode od 3 mL/min, brzini miješanja od 1000 rpm i koncentraciji polimera od 40 mg/mL te je postignuto zadovoljavajuće uklapanje melatonina. PEGilirane i nePEGilirane nanočestice s melatoninom u krutom stanju uspješno su pripremljene postupkom liofilizacije i predstavljaju potencijalno koristan terapijski sustav za liječenje bolesti usne šupljine.

Abstract (english)

The aim of this graduate thesis was the preparation of mucoadhesive and mucopenetrative polyanhydride nanoparticles loaded with melatonin. Melatonin is an endogenous compound that has shown significant therapeutic potential in the treatment of oral cavity diseases, immune system stimulation acting as an adjuvant to combat infections. Encapsulation of melatonin into the nanoparticles enhances its absorption through the mucus, avoiding the first passage metabolism, thus providing faster onset and localised action. Coating of the nanoparticle surface with polyethylene glycol can affect their transport through mucous. Therefore, we have prepared mucoadhesive (non-PEGylated) and mucopenetrative (PEGylated) nanoparticles. Technological process of nanoparticle preparation was optimised with the aim of preparing the nanoparticles of the targeted properties. The results show that by varying the concentration of polyhydric polymer is possible to control the size of nanoparticles, while the rate of addition of water and the mixing rate of the system during the desolvatation process is generally responsible for the formation of monodisperse nanoparticles. The nanoparticles of targeted properties were prepared at a water addition rate of 3 mL / min, a mixing rate of 1000 rpm and the polymer concentration of 40 mg / mL, with a satisfactory melatonin encapsulation efficiency. PEGylated and non-PEGylated melatonin nanoparticles in solid state are successfully prepared by lyophilization and represent a potentially useful delivery system for the treatment of oral cavity diseases.
