Abstract

Promjene u glikozilaciji, odnosno sijalilaciji humanog serumskog transferina događaju se kod brojnih akutnih i kroničnih bolesti te mogu biti jedan od pokazatelja stanja oboljenja. Razdvajanje različitih sijaloformi transferina korisno je za brojna znanstvena istraživanja i u dijagnostičke svrhe, ali često zahtijeva korištenje skupih analitičkih metoda. Ideja ovog rada bila je testirati kromatofokusiranje kao jeftiniju i jednostavniju metodu razdvajanja sijaloformi transferina za daljnje analize, uz naglasak na iskorištenje metode. Kromatofokusiranje s linearnim pH gradijentom pokazalo se kao metoda koja ima velik potencijal postati učinkovita i praktična metoda za analizu transferina i sličnih proteina, koja ima zadovoljavajuće iskorištenje i razdvajanje sijaloformi transferina.

Abstract (english)

Changes in glycosylation or sialylation of human serum transferrin occur in a number of acute and chronic diseases. Separation of various transferrin sialoforms is useful for numerous scientific studies and for diagnostic purposes but often requires the use of expensive analytical methods. The goal of this study was to test chromatofocusing as a cheaper and simpler method of separating transferrin sialoforms for further analysis, with emphasis on recovery. Chromatofocusing with a linear pH gradient has proven to be a method that has great potential in becoming an efficient and practical method for the analysis of transferrin and other glycoproteins, with sufficient recovery and satisfactory separation of protein sialoforms.
