Abstract | Analiza mokraće ima važnu ulogu kod dijagnoze i probira na bolesti te praćenju tijeka bolesti i uspješnosti
terapije. Određeni endokrinološki analiti zahtijevaju dodatak kiseline kao konzervansa u spremnik za
sakupljanje mokraće kako bi im stabilnost ostala očuvana do analize, dok se za određivanje biokemijskih
parametara ne mora sakupljati zakiseljeni uzorak. Kako se uzorak 24-satne mokraće najčešće sakuplja
tijekom vikenda, u ovom istraživanju ispitivan je utjecaj dodatka HCl na stabilnost proteina, albumina,
kreatinina, ureje, urata, kalcija, fosfata, magnezija, kalija, natrija, klorida, kortizola, VMA i 5-HIAA u
mokraći tijekom 24 i 48 sati kako bi se utvrdilo je li moguće u jednom zakiseljenom uzorku određivati
navedene analite čime bi se izbjeglo sakupljanje dva uzorka 24-satne mokraće (s i bez konzervansa).
Rezultati istraživanja pokazali su da su parametri kreatinin, ureja, urat, kalcij, fosfati, magnezij, kalij, natrij,
5-HIAA i VMA stabilni u zakiseljenom uzorku tijekom 24 i 48 sati. Kod određivanja koncentracije proteina
i albumina primijećeno je smanjenje koncentracije tijekom 24 i 48 sati, dok je kod određivanja kortizola i
klorida uočeno lažno povišenje koncentracije.
Iz dobivenih rezultata zaključujemo da ukoliko se uz katekolamine i metanefrine u mokraći traže pretrage
kreatinin, ureja, urat, kalcij, fosfati, magnezij, kalij, natrij, 5-HIAA i VMA, navedene pretrage mogu se
provesti iz jednog zakiseljenog uzorka 24-satne mokraće čime se smanjuje broj uzoraka koji dolaze u
laboratorij na analizu što olakšava pacijentima sakupljanje uzorka, a u laboratoriju se smanjuje mogućnost
pojave pogrešaka. |
Abstract (english) | Urine analysis plays an important role in the diagnosis and screening of diseases, as well as in monitoring the course of
diseases and the effectiveness of therapy. Certain endocrinological analytes require the addition of acid as a preservative
in the urine collection container to maintain their stability until analysis, while the collection of an acidified sample is
not necessary for the determination of biochemical parameters. Since the 24-hour urine sample is often collected over
the weekend, this study examined the effect of adding HCl on the stability of proteins, albumin, creatinine, urea, urate,
calcium, phosphate, magnesium, potassium, sodium, chloride, cortisol, VMA, and 5-HIAA in urine over 24 and 48 hours
to determine whether it is possible to measure these analytes in a single acidified sample, thus avoiding the collection
of two 24-hour urine samples (with and without preservatives).
The results of the study showed that creatinine, urea, urate, calcium, phosphate, magnesium, potassium, sodium, 5-
HIAA, and VMA are stable in acidified samples over 24 and 48 hours. However, a decrease in the concentration of
proteins and albumin was observed over 24 and 48 hours, while a false increase in the concentration of cortisol and
chloride was noted.
From the obtained results, we conclude that if tests for creatinine, urea, urate, calcium, phosphate, magnesium,
potassium, sodium, 5-HIAA, and VMA are required along with catecholamines and metanephrines in urine, these tests
can be performed from a single acidified 24-hour urine sample. This reduces the number of samples submitted to the
laboratory for analysis, making sample collection easier for patients and reducing the likelihood of errors in the
laboratory. |
