Abstract

E-NTPDaze su enzimska porodica koja obuhvaća mnoge ektoenzime uključene u metabolizam izvanstaničnih supstrata. Ekto-ATPaza pripada navedenoj enzimskoj skupini te je uključena u metabolizam nukleozid difosfata i nukleozid trifosfata. Smatra se da bi metabolizam navedenih molekula mogao imati ulogu u nastanku i razvoju mnogih patofizioloških procesa u organizmu s obzirom na njegovu sveprisutnost i važnost u većini tkiva. U ovom radu pokušalo se utvrditi postoji li statistički značajna razlika ekto-ATPazne aktivnosti između osoba oboljelih od kronične opstruktivne plućne bolesti i zdrave kontrolne populacije te može li se ista koristiti kao potencijalni biomarker za dijagnosticiranje i praćenje progresije bolesti. Aktivnost ekto-ATPaze određivala se pomoću Fiske-Subbarow metode koja je pokazala značajno variranje između izmjerenim vrijednostima. Izmjerena enzimska aktivnost iznosi 1.3849 ± 0.6058 U/L kod oboljelih od KOPB-a, a 1.1184 ± 0.2873 U/L kod zdravih osoba. Mann-Whitneyjev test je pokazao da ne postoji statistički značajna razlika u vrijednostima između ovih populacija Podaci su također statistički uspoređeni prema dobi, spolu i parametrima plućne funkcije. Utvrđeno je da ne postoji statistički značajna razlika između enzimske aktivnosti i dobi niti spola. S druge strane, pronađena je statistički značajna veza između ekto-ATPazne aktivnosti i parametara FEV1 i FEV1/FVC.

Abstract (english)

E-NTPDases are an enzyme family that includes many enzymes involved in metabolism of extracellular substrates. Ecto-ATPase is a member of this family which is responsible for hydrolysis of nucleoside triphosphates and nucleoside diphosphates.
The ubiquity of this enzyme suggests a possible involvement in numerous pathophysiological processes within the body. This assay attempts to establish the correlation between enzyme activities in a population of volunteers with chronic obstructive pulmonary disease and healthy control subjects. Furthermore it attempts to answer whether this method is adequate for diagnosis or progression monitoring of chronic obstructive pulmonary disease.
Ecto-ATPase activity was measured by Fiske-Subbarow method that exposed significant variation between measurements.
Results showed the average value of enzyme activity of 1.3849 ± 0.6058 U/L in population with COPD and 1.1184 ± 0.2873 U/L among healthy controls. No statistically significant difference was found between these groups using Mann-Whitney test.
Data was compared regarding sex, age and pulmonary function paramaters. No correlation was found between sex, age and ecto-ATPase activity. However, a statistically significant difference was found between ecto-ATPase activity and FEV1 and FEV1/FVC parameters.
