Abstract

Fitoestrogeni su spojevi biljnog podrijetla koji se u visokim koncentracijama nalaze u svakodnevnoj ljudskoj prehrani. Radi strukturne sličnosti sa 17-β- estradiolom pokazuju estrogene aktivnosti u biološkim testovima, te uzrokuju indukciju estrogenog učinka u sisavaca. Fitoestrogeni posjeduju niz korisnih učinaka za organizam, uključujući preventivno ili terapijsko djelovanje na karcinogenezu, aterosklerozu i osteoporozu. Budući da uvelike utječu na mehanizme u tijelu ovim radom se htjela ispitati njihova bioraspoloživost odnosno vezanje na proteine plazme. Korištena je brza gradijentna HPLC metoda na kiralnoj AGP koloni koja kao kiralni selektor ima imobilizirani α1-kiseli glikoprotein. Na osnovi provedenog istraživanja izračunate %AGP vrijednosti upućuju na širok raspon interakcija i postotka vezanja fitoestrogena sa α1-kiselim glikoproteinom. Korištena kromatografska metoda pokazala se prikladnom za ispitivanje vezanja fitoestrogena na α1-kiseli glikoprotein te su dobiveni rezultati prikladni za izvođenje kvantitativnih odnosa strukture i svojstava molekula.

Abstract (english)

Phytoestrogens are compounds of plant origin that are found in high concentrations in the daily human diet. Based on structural similarities with 17-β-estradiol they exhibit estrogenic activity in biological assays and cause the induction of the estrogenic effect in a mammal. Phytoestrogens possess a number of beneficial effects on the organism, including preventive or therapeutic effect on carcinogenesis, atherosclerosis and osteoporosis. Because they greatly affect the mechanisms in the body, this paper would examine their bioavailability and plasma protein binding. Fast gradient HPLC method was used on a chiral AGP column which has immobilized α1-acid glycoprotein for chiral selector. On the basis of the study, calculated % AGP values indicate a wide range of interactions and the percentage binding of phytoestrogens to α1-acid glycoprotein. Used chromatographic method proved to be suitable for testing binding of phytoestrogens to α1-acid glycoprotein and the obtained resultas are suitable for performing quantitative relationships between structure and properties of molecules.
