Abstract

Glikani su najrašireniji biopolimeri. Osim njihove uloge u metabolizmu, osnovne su sastavnice staničnih površina gdje su uključeni u vitalne stanične procese prepoznavanja. Zbog specifičnih interakcija s fiziološkim receptorima sudjeluju u mnogim biološkim procesima. Svi ti procesi su potencijalne mete terapeutskih intervencija.
Početak dvadesetog stoljeća obilježen je začetkom moderne medicine na čelu s terapijama baziranima na glikanima. Međutim, glikani su vrlo brzo postali zasjenjeni lakom dostupnošču terapija baziranih na DNA i proteinskim molekulama. Nedavni razvoj različitih alata i tehnika za analizu i sintezu strukturno složenih ugljikohidrata, ponovo je potaknuo interes terapeutske primjene glikana. Trenutno je mnogo glikanskih spojeva u različitim stadijima kliničkih ispitivanja. Ovaj diplomski rad je usmjeren na glikane kao farmaceutske mete, naglašava ulogu odobrenih glikanskih terapeutika te razmatra potencijal proteina koji mogu vezati ugljikohidrate. Rad također pruža uvid u cjepiva bazirana na ugljikohidratima, ona već razvijena ili u fazi razvoja, s posebnim naglaskom na anti-HIV te antitumorska sintetska cjepiva. U radu je isto tako opisana i važnost glikoinženjerstva proteina koji se upotrebljavaju u terapijama (npr. eritropoetina).

Abstract (english)

Glycans are the most abundant natural products. Besides their role in metabolism, they are fundamental constituents of every cell surface, where they are involved in vital cellular recognition processes. Because of their specific interactions with physiological receptors, they participate in many crucial biological processes. Therfore, there are many potential targets for therapeutic intervention. The beginning of the 20th century marked the dawn of modern medicine with glycan-based therapies at the forefront. However, glycans quickly became overshadowed as DNA- and protein-focused treatments became readily accessible. The recent development of new tools and techniques to study and produce structurally defined carbohydrates has spurred renewed interest in the therapeutic applications of glycans. Therefore, many compounds based on carbohydrates are now in various stages of clinical trials. This diploma thesis focuses on glycans as biopharmaceutical targets, it highlights the role of approved carbohydrate based drugs and discuss the potential of carbohydrate-binding proteins as new drug targets. It also presents carbohydrate based vaccines that have been or are being developed, with the emphasis on antitumoral sinthetic vaccines and anti-HIV vaccines, as well as glycoengineered proteins like erythropoietin.
