Abstract

CILJ ISTRAŽIVANJA: Cilj istraživanja je dobiti uvid u regulatorne zahtjeve za kontrolu onečišćenja u lijekovima kao i mogućnostima njihovog ispitivanja u lijekovima primjenom tehnike kapilarne elektroforeze. Naglasak bi bio stavljen na najveći izazov u određivanju onečišćenja u lijekovima tehnikom kapilarne elektroforeze, problem osjetljivosti.
MATERIJALI I METODE: U ovom teorijskom specijalističkom radu korištena je relevantna znanstvena i stručna literatura koja obrađuje problematiku ispitivanja onečišćenja u lijekovima. Korišteni su udžbenici, mrežne stranice te znanstvena i stručna literatura koja obrađuje tehniku kapilarne elektroforeze s posebnim naglaskom na određivanje onečišćenja u lijekovima i različitim načinima poboljšanja osjetljivosti metode.
RASPRAVA: Onečišćenje je svaki sastojak ljekovite tvari koji nema definiran kemijski entitet kao ljekovita tvar ili svaki sastojak farmaceutskog proizvoda koji nije ljekovita ili pomoćna tvar u samom proizvodu. ICH smjernice propisuju dopuštene granice za onečišćenja u farmaceutskim proizvodima.Za ispitivanje onečišćenja u lijekovima koristi se i kapilarna elektroforeza kao nova separacijska tehnika s nizom prednosti pred uobičajenim analitičkim tehnikama, poput on-line separacije, identifikacije i kvantifikacije analita, visoke učinkovitosti, niskih troškova održavanja, ekološke prihvatljivosti. Tehnika ima i neke nedostatke, posebice slabiju osjetljivost, što predstavlja izazov u analizi onečišćenja. Najčešće se problem osjetljivostiI riješava primjenom odgovarajućih kapilara, odabirom primjerenog detektora te on-line tehnikama ukoncentriravanja analita.
ZAKLJUČAK: Uz takve pristupe koji poboljšavaju osjetljivost metode kao najvećeg izazova u analizi onečišćenja, kapilarna elektroforeza svakako može biti tehnika izbora, komplementarna tehnici tekućinske kromatografije za ispitivanje onečišćenja u lijekovima.

Abstract (english)

OBJECTIVES: Objective of this research is to get an overview of regulatory demands on control of impurities in medicines, as well as possibilities on determination of impurities in pharmaceuticals by capillary electrophoresis. The emphasis would be on the biggest challenge in impurities determination by capillary electrophoresis, the issue of sensitivity.
MATERIALS AND METHODS: In this theoretical specialist thesis, relevant scientific and professional literature that deals with issues of impurities in pharmaceuticals was used. Textbooks, web pages, scientific and professional papers on capillary electrophoresis were also used, with special emphasis on impurities determination and different approaches on improvement of method sensitivity.
DISCUSSION: Impurity is any component of the new drug substance that is not the chemical entity defined as the new drug substance and any component of the new drug product that is not the drug substance or an excipient in the drug product. ICH guidelines prescribe limits for impurities in pharmaceutical products. For determination of impurities in pharmaceuticals, capillary electrophoreis is used as a new separation technique with numerous advantages over usual analytical techniques, such as on-line separation, identification and quantification of analytes, high efficiency, and low maintenance cost, ecological friendliness. Technique does have sensitivity challenges, which are addresses by the use of the appropriate capillary and detector, and different on-line preconcentration techniques.
CONCLUSION: With such approaches that enchance method sensitivity as the biggest challenge in determination of impurities, capillary electrophoresis can be the technique of choice, complementary to liquid chromatography technique for determination of impurities in pharmaceuticals.
