Abstract (english)

The main problem in cancer chemotherapy is non-selective action of cytotoxic drugs,
which leads to many side-effects. The problem could be solved by incorporation of drugs
into liposomes, or attaching them to polimers or antibodies. Monoclonal antibodies
against tumor selective/specific antigens have significant anticancer activity on their
own and are routinely used in the cancer therapy (rituximab, alemtuzumab, trastuzumab,
bevazicumab, cetuximab). Antibodies could also be used to target highly cytotoxic
drugs to tumor cells by preparing prodrugs, antibody-drug conjugates, which ensure
selective delivery and release of the cytotoxic drug within the cancer cell. Some experimental
therapies also include a prodrug of some cytotoxic drug in a combination with an
enzyme (ADEPT - antibody-directed enzyme prodrug therapy), an abzyme (ADAPT -
antibody-directed abzyme prodrug therapy) or selective gene delivery to the cancer cell
(GDEPT - gene-directed enzyme prodrug therapy).
