Abstract

Since the conventional anticancer drugs encounter many obstacles, nanotechnology offers a wide range of new possibilities among which are polymersomes (Ps) as potential drug delivery systems. The aim of this study was to prepare and characterize Ps made out of poly(dimethylsiloxane)-block-poly(2-methyloxazoline) (PDMS-b-PMOXA) diblock copolymer, to test their stability and finally their behaviour towards two different breast cancer cell lines. The results of the study included size and polydispersity index (PDI) of the nanosystems determined by using dynamic light scattering (DLS). Furthermore, imaging of the particles was done using transmission electron microscopy (TEM) while the stability was tested in two different buffers relevant for the study. In vitro experiments, in particular cell viability assay, were done on MDA-MB-231 and MCF-7 breast cancer cell lines.

Abstract (croatian)

Nanotehnologija može značajno pridonijeti razvoju antitumorskih lijekova. Uklapanjem antitumorskih lijekova u odgovarajuće terapijske nanosustave kao što su polimerosomi može se povećati njihova djelotvornost i/ili sigurnost. Cilj je ovog diplomskog rada bio priprema i karakterizacija polimerosoma građenih od poli(dimetilsiloksan)-blok-poli(2-metiloksazolin) (PDMS-b-PMOXA) diblok kopolimera, ispitivanje njihove stabilnosti i biokompatibilnosti. Veličina čestica te indeks polidisperznosti (PDI) određeni su metodom raspršenja laserske svjetlosti, a oblik i morfologija polimerosoma metodom transmisijske elektronske mikroskopije. Stabilnost polimerosoma ispitana jr koristeći dva različita pufera. Od in vitro pokusa provedeo je ispitivanje biokompatibilnosti koristeći MDA-MB231 i MCF-7 stanične linije.
