Abstract

Šećerna bolest je metabolički poremećaj kojeg karakterizira hiperglikemija, poremećaj u metabolizmu
ugljikohidrata, masti i proteina koji dovodi do manjkave sekrecije i/ili aktivnosti inzulina. Smatra se da oksidativni
stres ima ključnu ulogu u nastanku i razvoju šećerne bolesti, kao i njenih kasnijih komplikacija.Tretiranje HepG2
stanica 20 mM glukozom stvara hiperglikemijske uvjete koji dovode do oksidativnog stresa, što se očituje značajnim
padom aktivnosti enzima glutation peroksidaze (GPx) i glutaton S-transferaze (GST) (p < 0.05).
Cilj ovog rada bio je ispitati antioksidativni učinak vrsta roda Globularia na aktivnost antioksidativnih enzima GPx i
GST te ga povezati s kemijskim sastavom četiriju ispitivanih vrsta roda Globularia: (G. alypum L., G. cordifolia L.,
G. meridionalis (Podp.) O,. Schwarz L. i G. punctata Lapeyr.). Aktivacijom enzima GPx od strane vrsta roda
Globularia dokazan je antioksidativni učinak. Aktivnosti enzima izmjerene su u lizatu stanica. U istraživanju je
uočeno statistički značajno smanjenje aktivnosti GPx u lizatu HepG2 stanica tretiranih 20 mM glukozom (D) u
odnosu na kontrolnu skupinu (C) čime je dokazan oksidativni stres do kojeg je dovela glukoza.Rezultati ovog rada
upućuju na antioksidativni učinak vrsta roda Globularia (0.5 i 1.0 mg/ml) jer je nakon tretiranja došlo do povećane
aktivnosti GPx enzima u HepG2 stanicama u hiperglikemijskim uvjetima. Između četiri proučavana ekstrakta, dvije
vrste (G. punctata Lapeyr. i G. cordifolia L.) su se istaknule statistički značajnim porastom aktivnosti
antioksidativnih enzima GPx i GST (p < 0.05). Antioksidativno djelovanje spomenutih vrsta pripisujemo
najvjerojatnije flavonoidima i iridoidima, spojevima čije prisustvo je u njima dokazano, a čija je aktivnost u
znanstvenoj literaturi opisana.Kako bismo potvrdili naše zaključke mogla bi se provesti daljnja istraživanja in vivo
čime bi se potvrdio njihov antioksidativni učinak.

Abstract (english)

Diabetes mellitus is metabolic disorder associated with hyperglycemia, disturbance in carbohydrates, protein and fat
metabolism resulting with lack of secretion and/or activity of insulin. It is considered that oxidative stress has pivotal
role in the etiology and pathogenesis of the diabetes mellitus, as well as its late complications.
Treatment of HepG2 cells by 20 mM glucose induces hyperglycemic condition, leading to oxidative stress which is
manifested with significant decrease of GPx and GST enzyme activity (p < 0.05). Intent of this study is to investigate
antioxidative effect of Globularia spp on activity of antioxidative enzymes GPx and GST, and relate it with chemical
composition of four tested plant species : (G. alypum L., G. cordifolia L., G. meridionalis (Podp.) O,. Schwarz L. i
G. punctata Lapeyr.). Activation of the GPx enzyme by Globularia species has proven their antioxidative effect.
Statistically significant decrease of GPx activity, in HepG2 cells lysates treated by 20 mM glucose (D) compared to
control group (C), is observed. Mentioned species authenticated oxidative stress which was caused by glucose.
Activities of these enzymes were measured in cell lysates.
The results of this study imply the antioxidative effect of Globularia spp (0.5 and 1.0 mg/mL), since it led to
enhanced activity of GPx enzyme after treating in HepG2 cells in hyperglycemic condition. Among four tested plant
extracts, two species (G. punctata Lapeyr. i G. cordifolia L.) are highlighted with its significant increase of enzyme
activity of antioxidative enzymes GPx and GST (p < 0.05). Antioxidative effect among mentioned species probably
attributes to flavonoides and iridoides, chemical compounds with proven presence, which activity is described in
relevant science literature. Additional in vivo studies are required to substantiate our conclusions.
